- Sermorelin is a synthetic analogue of the first 29 amino acids of endogenous GHRH [GHRH(1-29)-NH2]; it stimulates endogenous pituitary GH secretion by binding pituitary GHRH receptors — unlike exogenous somatropin, it preserves the hypothalamic-pituitary feedback axis and physiological pulsatile GH secretion patterns.
- The Geref International Study Group multicenter trial (JCEM 1996, n=110 GHD children) demonstrated that once-daily subcutaneous sermorelin 30 µg/kg increased mean height velocity from 4.1 to 8.0 cm/year after 6 months; however, a direct comparison study (PMID 8329830) confirmed somatropin achieved faster growth (14.6 vs ~9 cm/year), establishing sermorelin as a less potent but mechanistically distinct alternative.
