What is Pancragen? Research, safety, and status

Pancragen is a four-amino-acid peptide: lysine, glutamic acid, aspartic acid, tryptophan, with an amide cap on the C-terminus. Khavinson's 2005 paper explains the design. Comparing the sequences of insulinotropic (insulin-releasing) polypeptides, the group found a shared four-residue fragment, then synthesized "KEDWa," a version protected from gastrointestinal proteases. The "a" is the amide.

The proposed mechanism follows the group's general model. The peptide is said to reach the nucleus and bind DNA. The 2005 paper suggested it switches on the preproinsulin gene by pairing with GGCAGG/CCTGCC sequences. A 2014 molecular-mechanics paper proposed GGCAG as the binding site, with contacts in both the major and minor grooves of DNA. A 2015 paper reported that KEDW changed DNA methylation at a few CpG sites in the FOXA2 promoter of cultured pancreatic cells, but the authors found no correlation with that gene's expression. All of this is modeling or cell culture from the developer group.

Every study we found has Khavinson as an author or names his institute as the source of the peptide. No independent lab has replicated the glucose effects.

The developer literature describes the C-terminal amide. One online peptide database lists 576 Da, which fits the free acid instead. On a COA, that one-dalton difference matters.
